※ Avibactam (NXL-104), A diaza dicycloctanone compound, is the most promising non-β -lactamase inhibitor. Although it has no obvious antibacterial activity, it can inhibit β -lactamase of type A (including ESBL and KPC) and type C. Therefore,when used in combination with various cephalosporins and carbapenems, has a broad spectrum of antimicrobial activity, particularly against EScherichia coli and Klebsiella pneumoniae that contain ultra broad spectrum β -lactamase, escherichia coli that contain excessive AmpC enzyme, and escherichia coli that contain both AmpC and ultra broad spectrum β -lactamase.

※ β -lactamase serine nucleophilic attack avebatamide bond, ring-opening to form covalent conjugate, enzyme - inhibitor complex, enzyme inhibition form, and no hydrolysis, and then through cyclization to form lactam ring and avebatamide. The rate of ring-opening was much higher than that of cyclization due to nucleophilic attack, and β - lactamase was basically inhibited. In this process, the structure of avitabtam can be restored by reverse reaction, so it has a long-term inhibitory effect on enzyme. Avibactam is a novel β -lactamase inhibitor with a non-lactamase scaffold that irreversibly inhibits the lactamase of Mycobacterium tuberculosis.
