    Structural features: The glycosyl skeleton is composed of 1',2' -dideoxyribose, the hydroxyl group at the 5' position is protected by dimethoxy-triphenylmethyl (DMT), and the 3' position is connected with a phosphoramidine group, including two substituents: 2-cyanoethyl and N, n-diisopropyl. This structure enables the reagent to specifically introduce a stable abasic site in oligonucleotide synthesis.

    Function and Application 1

    Introducing baseless sites: A single glycosylated spacer can be introduced inside the oligonucleotide or at the 5' end to simulate baseless sites. This is very useful in the processes of studying DNA damage, repair and gene mutations, for example, it is used to study mutations caused by depurination.

    Reducing interactions: As a spacer, it can increase the distance between oligonucleotides and modifying groups, reducing the possible adverse interactions between modifying groups and oligonucleotide sequences. For instance, for sequences rich in G, the use of this reagent can separate modifying groups such as fluorescent dyes from oligonucleotides, reducing the fluorescence quenching effect.

    Synthetic application 3: During the oligonucleotide synthesis process, its usage method is similar to that of standard phosphoramidite reagents. The coupling and deprotection steps usually do not require changes to the standard method recommended by the synthesizer manufacturer.

    Properties and Stability 3: It is usually solid and needs to be stored under dry conditions. It is generally recommended to store it in a frozen environment at -10 ℃ to -30 ℃. Its stability is approximately 2 to 3 days.
