Edman-N sequencing

Protein biosynthesis starting from the N-terminal, the amino acid sequence of N-terminal has a strong impact on the biological functions, such as the half-life of protein, the sub cellular localization of protein. Certain posttranslational modifications involving the protein function and the stability also occur on N-terminal. Therefore by analyzing the N-terminal sequence, the person can analyze the spatial structures of proteins and evaluate the biological function of proteins.

With the development of modern pharmaceutical industry, there have been a lot of protein and polypeptide drugs molecules. Analyzing the N-terminal amino acid sequence is a critical link in pharmaceutical quality control.

At present, there are two main methods to study the protein n-terminal sequence -non-mass spectrum technology, such as traditional Edman degradation, RT-PCR reverse transcription, and mass spectrometry. Each method has its own advantages and shortcomings. The technology based on the traditional Edman degradation has become more common on the market.
